6. Abstract:

 

High-throughput data from various omics screens are providing large numbers of candidate genes involved in various cellular processes. Their candidacy however needs to be validated by a series of relevant bench experiments. Several such efforts carried out in our laboratory will be presented. The first topic is about identifying direct regulatory targets of transcription factors. Sox10 and Sox2 are two transcription factors that respectively play key roles in maintaining unique properties of Schwann cells and neural stem cells. How differing strategies were applied in search for their direct regulatory targets will be discussed. The second topic concerns isolating tumor suppressors and oncogenes from large sets of high-throughput data. We have recently carried out exomeseq and RNAseq using samples from lung adenocarcinoma and angioimmunoblastic T cell lymphoma patients. In vitro and in vivo experiments are underway to confirm their activity in carcinogenesis and latest results will be presented.
